Q8(R2): Pharmaceutical Development

Holm, Per, Allesø, Morten, Bryder, Mette C., Holm, René·2017·ICH Quality Guidelines

Abstract

International Conference on Harmonisation (ICH) Q8(R2) provides guidance on the contents of the Pharmaceutical Development section in regulatory submissions, that is, new drug applications (NDAs) or marketing authorization applications (MAAs). The guideline thereby proposes a new approach both in pharmaceutical development and subsequently in quality control (QC) within the commercial manufacturing arena, as the development process is now specifically designed to support it. Pharmaceutical companies are therefore encouraged to apply a systematic, scientific approach to drug development. This chapter describes the two development paradigms (quality by testing (QbT) and quality by design (QbD)) that contain many of the same elements, and focuses specifically on those elements important to a QbD approach. It exemplifies how rational selection criteria for choosing process technology can be defined based on public and prior knowledge. In addition, the proposal for a manufacturing classification system (MCS) provides additional insight, including the use of appropriate expert systems.

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
