Thioflavin Derivatives as Markers for Amyloid‐β Fibrils: Insights into Structural Features Important for High‐Affinity Binding

Leuma Yona, Rodrigue, Mazères, Serge, Faller, Peter, Gras, Emmanuel·2008·ChemMedChem

Abstract

How do we go together? Structural variations have been used to obtain information regarding the interaction of thioflavin-derived markers and amyloid-β fibril targets. Affinity variations of up to three orders of magnitude were observed, revealing some structural requirements for these markers at the molecular level.

This paper’s full text isn’t openly available, so its claims and self-check weren’t run. The registry and citation checks still apply.

Claims other papers make about this one

Each is checked against this paper’s own text: does it say what the citing paper says it does?

  • Titrations with various amounts of compounds reveal competitive binding curves that yield Ki values 180±25 nM and 36±6 nM for L1 and L2, respectively.

“Titrations with various amounts of compounds reveal competitive binding curves that yield Ki values 180±25 nM and 36±6 nM for L1 and L2, respectively (Figure 6b).84,85”
  • Performing the same titration of Aβ fibrils with ThT yields a Kd of 1.17±0.14 μM, a value similar to those reported previously.

“By comparison, performing the same titration of Aβ fibrils with ThT yields a Kd of 1.17±0.14 μM (Figure S21b), a value similar to those reported previously.83,85”
  • L1 exhibits a high affinity for Aβ fibrils comparable to other neutral ThT derivatives, suggesting that appending a metal-binding group to the 2-phenylbenzothiazole fragment does not limit its amyloid binding affinity.

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
