Abstract

The plasma membrane (PM) is composed of a complex lipid mixture that forms heterogeneous membrane environments. Yet, how small-scale lipid organization controls physiological events at the PM remains largely unknown. Here, we show that ORP-related Osh lipid exchange proteins are critical for the synthesis of phosphatidylinositol (4,5)-bisphosphate [PI(4,5)P 2 ], a key regulator of dynamic events at the PM. In real-time assays, we find that unsaturated phosphatidylserine (PS) and sterols, both Osh protein ligands, synergistically stimulate phosphatidylinositol 4-phosphate 5-kinase (PIP5K) activity. Biophysical FRET analyses suggest an unconventional co-distribution of unsaturated PS and phosphatidylinositol 4-phosphate (PI4P) species in sterol-containing membrane bilayers. Moreover, using in vivo imaging approaches and molecular dynamics simulations, we show that Osh protein-mediated unsaturated PI4P and PS membrane lipid organization is sensed by the PIP5K specificity loop. Thus, ORP family members create a nanoscale membrane lipid environment that drives PIP5K activity and PI(4,5)P 2 synthesis that ultimately controls global PM organization and dynamics.

  • PI(4,5)P2 clusters in the cytoplasmic leaflet align with cholesterol- and sphingomyelin-rich regions in the external leaflet by trans-bilayer coupling.

“Basically, current studies strongly suggest that PI(4,5)P2 clusters in the cytoplasmic leaflet align with cholesterol- and sphingomyelin-rich regions in the external leaflet of the plasma membranes by a mechanism referred to as trans-bilayer coupling [111,127–130].”

From Phosphatidylinositol(4,5)bisphosphate: diverse functions at the plasma membrane (2020) · cites this paper as [130]

The Results passages on lipid co-distribution and the PIP5K specificity loop describe PI4P, phosphatidylserine, and cholesterol interactions, not alignment of PI(4,5)P2 clusters with regions in the external leaflet. Searches for trans-bilayer coupling and sphingomyelin found no passage in our copy addressing the claimed alignment. (LLM, conf=0.76)

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
