Abstract

Significance: Vitamin B 12 is required by humans and a variety of other organisms for diverse metabolic processes, but is produced only by a subset of microorganisms. The anaerobic biosynthesis of the “lower ligand” of B 12 , 5,6-dimethylbenzimidazole (DMB), is the only unknown component of the B 12 biosynthetic pathway. We report the identification of the bzaABCDE genes that are necessary and sufficient for the anaerobic biosynthesis of DMB. We have characterized the role of each of the bza genes and identified three intermediates in the pathway. This finding not only completes the B 12 biosynthesis pathway but also enables the sequence-based prediction of cobamides synthesized by anaerobic microorganisms.

Claims other papers make about this one

Each is checked against this paper’s own text: does it say what the citing paper says it does?

  • The anaerobic synthesis of DMB is performed by the bzaABCDE gene products, starting from AIR.

“The anaerobic synthesis of the cobalamin lower ligand dimethylbenzimidazole (DMB) is performed by the bzaABCDE gene products [39], starting from 5‐aminoimidazole ribotide (AIR), an intermediate of thiamine and purine biosynthesis.”
  • In some organisms, only bzaF is present, generating HBI as the cobamide lower ligand.

“In some organisms, only the product of bzaF, a radical SAM enzyme homologous to bzaA, bzaB and thiC products, is present [39, 109], generating 5‐hydroxybenzimidazole (HBI) as the cobamide lower ligand (Fig. 1C).”
  • Among enzymes involved in synthesis of cobalamin lower ligand, only BzaF is well conserved across the bacterial-archaeal divide.

“Among the enzymes involved in the synthesis of the cobalamin lower ligand, only BzaF, which catalyzes the synthesis of 5‐hydroxybenzimidazole (HBI) [39], is well conserved across the bacterial–archaeal divide”

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
