Abstract

Different vesicular cargoes have distinct responses to tau phosphorylation. Phosphomimetic (Y18E) tau has a weakened effect on lysosome motility compared with unphosphorylated tau and preferentially inhibits peripheral lysosomes. In contrast, early endosomes are strongly inhibited by both unphosphorylated and phosphomimetic tau.

  • Mutations at Y18 increase tau dynamics on microtubules, reduce tau accumulation, and impair tau-mediated regulation of intracellular transport.

“mutations at Y18, the terminal residue of PAD, increase tau dynamics on microtubules, reduce tau accumulation, and impair tau-mediated regulation of intracellular transport”

From Tau hyperphosphorylation impairs cooperative binding to microtubules and perturbs organelle trafficking in neurons (2026) · cites this paper as Balabanian et al., 2022

The Introduction (page unavailable) reports that phosphomimetic Y18E tau has reduced microtubule affinity and increased diffusivity, while the Discussion proposes that a negative charge at Y18 may favor dynamic microtubule binding. The Results (page unavailable) show reduced tau-mediated inhibition of lysosome transport at low Y18E expression, but stronger inhibition of early-endosome motility; the effect on transport is therefore cargo-dependent. Our copy does not establish reduced tau accumulation, and it presents the proposed change in binding dynamics more cautiously than the claim. (LLM, conf=0.86)

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
