Some gating potentiators, including VX-770, diminish ΔF508-CFTR functional expression
Open accessAbstract
Ivacaftor, a potentiator of ΔF508-CFTR channel function in cystic fibrosis, reduces the ability of corrector drugs to rescue the ΔF508-CFTR membrane trafficking defect.

Chronic treatment with 1 μM VX-770 significantly decreased functional correction by VX-809 in the presence of acute potentiator addition.
“Chronic treatment with 1 μM VX-770, a concentration that is therapeutically reached in respiratory epithelia (34), was reported to significantly decrease the functional correction by VX-809 in the presence of acute potentiator addition (Figure 3, A and B), as observed in CF-HBE (35, 36) and CF-HNE (21).”
From Allosteric folding correction of F508del and rare CFTR mutants by elexacaftor-tezacaftor-ivacaftor (Trikafta) combination (2020) · cites this paper as [35]
The Results section on chloride conductance reports that 24 hours of VX-770 reduced VX-809–corrected ΔF508-CFTR current in primary HBE cells, with current measured after acute potentiator addition. But the reported chronic dose for those functional measurements was 100 nM, not the claimed 1 μM; our copy mentions 1 μM in a different, pulse-chase experiment (Materials and Methods). No page numbers are available in our copy. (LLM, conf=0.88)

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
