Synthetic lethality in KRas-driven cancer cells created by glutamine deprivation
Open accessAbstract
deaminated to glutamate by glutaminase (GLS).Glutamate is then converted to α-ketoglutarate by glutamate oxaloacetate transaminase (GOT).Aminooxyacetate (AOA) inhibits GOT and therefore suppresses generation of α-ketoglutarate from Gln-derived glutamate. The aspartate generated by GOT is critical for nucleotide and amino acid biosynthesis.B. The relative positions of the growth factor-dependent Restriction Point and late G 1 metabolic checkpoints mediated by essential amino acids (EAA), Gln and mTOR are depicted.

KRAS-dependent upregulation of GOT1/2 in pancreatic cancer facilitates aspartate production for nucleotide biosynthesis and allows NADPH generation via ME1.
“For instance, KRAS-dependent upregulation of Glutamate Oxaloacetate Transaminase 1,2. (GOT) expression in pancreatic cancer facilitates production of aspartate for nucleotide biosynthesis, and allows NADPH generation via ME157,58 (Fig 2).”

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
