Charge Transfer at the Qo-Site of the Cytochrome bc1 Complex Leads to Superoxide Production

Salo, Adrian Bøgh, Husen, Peter, Solov’yov, Ilia A.·2017·The Journal of Physical Chemistry B

Abstract

The cytochrome bc 1 complex is the third protein complex in the electron transport chain of mitochondria or photosynthetic bacteria, and it serves to create an electrochemical gradient across a cellular membrane, which is used to drive ATP synthesis. The purpose of this study is to investigate interactions involving an occasionally trapped oxygen molecule (O 2 ) at the so-called Q o site of the bc 1 complex, which is one of the central active sites of the protein complex, where redox reactions are expected to occur. The investigation focuses on revealing the possibility of the oxygen molecule to influence the normal operation of the bc 1 complex and acquire an extra electron, thus becoming superoxide, a biologically toxic free radical. The process is modeled by applying quantum chemical calculations to previously performed classical molecular dynamics simulations. Investigations reveal several spontaneous charge transfer modes from amino acid residues and cofactors at the Q o -site to the trapped O 2 molecule.

Claims other papers make about this one

Each is checked against this paper’s own text: does it say what the citing paper says it does?

  • It seems unlikely that coherence in biological systems extends beyond picoseconds for photosynthesis and milliseconds for the radical pair mechanism.

“While research suggests that long-lived coherence in photosynthetic systems lasts for picoseconds 194 and the lifetime of the radical pair mechanism is discussed in terms of milliseconds 161 , it seems unlikely that coherence in biological systems extends beyond these timescales.”

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
