APOE3 Christchurch Heterozygosity and Autosomal Dominant Alzheimer’s Disease

J Nicholas Cochran, Michael D. Greicius, Alison Goate·2024·New England Journal of Medicine

Abstract

To the Editor: The report by Quiroz et al. (June 20 issue) 1 on the Colombian PSEN1 E280A pedigree suggests a protective effect of apolipoprotein E3 Christchurch ( APOE3 Ch ) variant heterozygosity. The signal from 27 carriers is driven by 13 carriers in whom mild cognitive impairment or dementia developed. A previous study assessed 11 of these 13 heterozygotes with the use of age-at-onset data systematically collected before knowledge of the APOE3 Ch variant; the median age at onset of mild cognitive impairment was 1 year earlier in APOE3 Ch carriers than in noncarriers, and the median age at onset of dementia was equal in carriers . . .

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
