Backwaters and rapids on the amyloid river
Abstract
The advent of amyloid imaging1 has ushered in a new era for the field of Alzheimer disease (AD), opening the door for approaching previously unapproachable questions. No longer are researchers restricted to postmortem measurement of the neuropathologic hallmark of AD—amyloid plaques. Now, researchers are able to detect the presence and burden of amyloid in the living brain and examine how amyloid burden changes over time.

Plaque deposition essentially occurs during the run-up to clinical AD.
“Recent data indicate plaque deposition essentially occurs during the run-up to clinical AD [216, 217].”

“It is also important to consider, however, that if amyloid turns out to be only a secondary biomarker of the disease, as opposed to its cause, then targeting its accumulation at any stage will have, at most, a limited effect on clinical outcomes.(7) Finally, the fibrillar deposition of amyloid in the brain parenchyma…”

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
