Optimization of in vitro and ex vitro regeneration and micromorphological studies of Micrococca mercurialis (L.) Benth
Open accessAbstract
A regeneration protocol has been successfully developed using nodal segments and leaf explants of Micrococca mercurialis (L.) Benth. The surface sterilized leaf explants were induced to proliferative green colored callus on Murashige and Skoog (MS) me dium supplemented with 0.5 mg l -1 of 2,4-dichlorophenoxyacetic acid (2,4-D).Shoots were regenerated from the callus on MS medium sup plemen ted with 0.5 mg l -1 6-benzylaminopurine (BAP).The nodal shoot segments res ponded on MS medium augmented with 1.0 mg l -1 BAP.The shoots were mul ti pliedonMSmediumfortifiedwith0.5mgl-1 each of BAP, kinetin (Kin) and 0.1 mg l -1 α-naphthaleneaceticacid(NAA).Themaximum17.3shootswith8.58 cm average length were resulted on this media combination. The shoots were rooted ex vitro when pulse treated with 100 mg l -1 indole-3 butyric acid (IBA).Theplant letsweresuccessfullyestablishedinthefieldwith96%success rate.Mic ro mo rphological studies were conducted for the successive stages of in vitro, ex vitro and in vivo grown plants to analyze the foliar developmental changes taken place across the environments. It has been concluded that the abnormal mor phophysiology of in vitro plantlets were repaired while acclimatization under thegreenhouseconditionsbeforetransferringtothefield, whichspeedupthe micropropagation process and increase the survival rate of the plantlets.K e y w o r d s : Micrococca mercurialis, regeneration, ex vitro rooting, micromorphological studies, organogenesis Р Е З Ю М Е Манокари М., Шехават М.С.Оптимизация процесса регенерации in vitro и ex vitro и микроморфологические исследования Micrococca mercurialis (L.) “When producing Persian walnut in vitro explants, it is crucial to undergo hardening and acclimatization processes. This is due to the dysfunctional stomata of walnut in vitro-plantlets, resulting in excessive transpiration and heightened sensitivity to environments with increased evaporative demands and solar radiation (Chandra et al., 2010; Rathore et al., 2013; Manokari and Shekhawat, 2017).”

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
