Role of Metal Availability in Modulating Antibiotic Efficacy
Open accessAbstract
Understanding how host factors influence antibiotic efficacy is critical to preserving or enhancing treatment efficacy. The limitation of trace metals is a key aspect of the immune response, with the infection microenvironment depleted of essential metals through the action of host immune proteins. Here, we demonstrate that the limitation of zinc and manganese has opposing effects on β-lactam antibiotic efficacy against Staphylococcus aureus. Our results show that zinc chelation by the host immune protein calprotectin directly inactivates bacterial autolysins, zinc-dependent enzymes required for cell wall degradation and bacterial lysis following β-lactam treatment. Comparison of antibiotic efficacy in the presence and absence of calprotectin, both in vitro and in vivo, shows significantly decreased antibiotic killing under conditions of zinc limitation. On the other hand, in zinc-replete environments, manganese limitation was found to potentiate β-lactam antibiotic killing. Our data suggests that bacterial adaptation to intracellular oxidative stress under Mn-limiting conditions results in the increased expression and activity of bacterial autolysins, ultimately sensitizing S. aureus to antibiotic treatment. These findings demonstrate how zinc and manganese availability can dramatically influence the outcome of antibiotic treatment against S. aureus, highlighting a previously unrecognized determinant of antibiotic susceptibility for β-lactam antibiotics within the host.

TL;DR
In *Staphylococcus aureus*, host calprotectin-mediated zinc limitation inactivates zinc-dependent autolysins and induces tolerance to β-lactam antibiotics, whereas manganese limitation in zinc-replete conditions increases autolysin expression/activity and potentiates β-lactam killing. The thesis reports in vitro and murine bacteremia evidence that metal availability is a determinant of β-lactam efficacy.
Key findings
- Calprotectin (CP) increased *S. aureus* survival against β-lactams in a dose-dependent manner; 60 µg/mL CP yielded approximately 50-fold more survivors after oxacillin and nafcillin treatment, with no change in MIC and no effect on ciprofloxacin efficacy.
- Zinc limitation drove β-lactam tolerance: CP variants lacking both metal-binding sites failed to induce tolerance, TPEN caused up to a 1000-fold increase in survivors, ZnSO₄ restored antibiotic efficacy, and CP inhibited Atl autolytic activity; Δatl abolished CP-induced oxacillin tolerance. In a murine bacteremia model, oxacillin-treated CP-deficient S100A9⁻/⁻ mice had approximately 20-fold fewer bacteria in the liver than wild-type mice.
- Manganese limitation potentiated β-lactam killing: ΔmntH/C in Mn-deplete medium showed increased oxacillin killing; autolytic activity was approximately 40% higher for ΔmntH/C and approximately 20% higher for wild-type under Mn-deplete conditions. Wild-type CP or ΔS2 CP increased Atl and Sle1 expression around 4-fold, and methyl viologen at 50 µM potentiated while 1 mM reduced oxacillin efficacy.
Methods
- In vitro assays used *S. aureus* Newman, JE2, and clinical MSSA/MRSA isolates grown in TSB-based or Chelex-treated TSB with defined metals; timed kill curves with β-lactams plus CP, CP site mutants, TPEN, EGTA, BSA, ZnSO₄, or methyl viologen; CFU plating, MIC determination, autolysis assays with heat-killed RN4220, zymography, HADA/WGA fluorescence microscopy, TEM, ADP/ATP measurement, and RT-qPCR.
- In vivo murine bacteremia model: wild-type and S100A9⁻/⁻ C57BL/6 mice were infected retro-orbitally with 10⁷ CFU *S. aureus* Newman; after 48 hours, oxacillin was given intraperitoneally at 400 mg/kg every 12 hours for three days, and organs were harvested at day five for bacterial burden.
- Statistical analyses included Kruskal-Wallis with Dunn’s test, Mann-Whitney test, Welch’s ANOVA with Dunnett’s T3, one-way ANOVA with Tukey’s test, and Welch’s T-test, with significance set at p≤0.05.
Limitations
- The paper states that CP is not the only zinc-binding protein at infection sites; S100A7, S100A12, and other zinc-restriction mechanisms could contribute, and CP’s contribution varies by tissue. In vivo, oxacillin efficacy was significantly improved in the liver but not in kidney or heart.
- The paper states that CP-induced tolerance at lower concentrations is not simply due to slowed growth, but this does not eliminate possible growth-delay contributions in a subset of cells; at higher CP concentrations, other zinc- and manganese-dependent processes may also affect tolerance.
- The paper states that the exact mechanism by which ROS modulate autolytic activity is unknown and that manganese’s effect is indirect, likely through regulatory pathways; the text also makes evident that Chapter 3 is intended for a future manuscript and some expression experiments used N=2.

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Claims read by deepseek-v4.1-flash under definition version 3 (extract-v3) on 2026-11-05.
Classified by jev-1.13-20260917 (gold-guideline-v0.2) on 2026-11-05.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
