Abstract

Summary: Mounting evidence suggests that mitochondrial dysfunction plays a causal role in the etiology and progression of Alzheimer's disease ( AD ). We recently showed that the carbonic anhydrase inhibitor ( CAI ) methazolamide ( MTZ ) prevents amyloid β (Aβ)‐mediated onset of apoptosis in the mouse brain. In this study, we used MTZ and, for the first time, the analog CAI acetazolamide ( ATZ ) in neuronal and cerebral vascular cells challenged with Aβ, to clarify their protective effects and mitochondrial molecular mechanism of action. The CAI s selectively inhibited mitochondrial dysfunction pathways induced by Aβ, without affecting metabolic function. ATZ was effective at concentrations 10 times lower than MTZ . Both MTZ and ATZ prevented mitochondrial membrane depolarization and H 2 O 2 generation, with no effects on intracellular pH or ATP production. Importantly, the drugs did not primarily affect calcium homeostasis. This work suggests a new role for carbonic anhydrases ( CA s) in the Aβ‐induced mitochondrial toxicity associated with AD and cerebral amyloid angiopathy ( CAA ), and paves the way to AD clinical trials for CAI s, FDA ‐approved drugs with a well‐known profile of brain delivery.

From Proteogenomic analysis of human cerebrospinal fluid identifies neurologically relevant regulation and implicates causal proteins for Alzheimer’s disease (2024) · cites this paper as [90]

The Introduction identifies acetazolamide as a carbonic anhydrase inhibitor, and the Discussion reports that it reversed Aβ-induced mitochondrial depolarization and H2O2 production in the study’s cell models. The passages identify other carbonic anhydrase isoforms but do not establish that acetazolamide targets CA12 specifically. Page numbers were not supplied. (LLM, conf=0.88)

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
