The interaction between progranulin and prosaposin is mediated by granulins and the linker region between saposin B and C
Open accessAbstract
The frontotemporal lobar degeneration (FTLD) protein progranulin (PGRN) is essential for proper lysosomal function. PGRN localizes in the lysosomal compartment within the cell. Prosaposin (PSAP), the precursor of lysosomal saposin activators (saposin A, B, C, D), physically interacts with PGRN. Previously, we have shown that PGRN and PSAP facilitate each other's lysosomal trafficking. Here, we report that the interaction between PSAP and PGRN requires the linker region of saposin B and C (BC linker). PSAP protein with the BC linker mutated, fails to interact with PGRN and deliver PGRN to lysosomes in the biosynthetic and endocytic pathways. On the other hand, PGRN interacts with PSAP through multiple granulin motifs. Granulin D and E bind to PSAP with similar affinity as full‐length PGRN. image Read the Editorial Comment for this article on page 154.

Prosaposin heterodimerizes with progranulin, regulating its levels and facilitating its transport to the lysosome.
“Prosaposin heterodimerizes with progranulin, regulating its levels and facilitating its transport to the lysosome35,36.”
From Rare variant associations with plasma protein levels in the UK Biobank (2023) · cites this paper as [35]
The Abstract reports that prosaposin physically interacts with progranulin and helps target it to lysosomes. The Introduction says PSAP polymorphisms affect plasma progranulin levels. The paper describes a complex, but the cited passages do not establish that it is specifically a heterodimer. (LLM, conf=0.83)

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
