Identification of DNA elements involved in somaclonal variants of Rauvolfia serpentina (L.) arising from indirect organogenesis as evaluated by ISSR analysis

Open access

Abstract

Rauvolfia serpentina an endangered medicinal plant was chosen for in vitro propagation using modified MS medium. Genetic fidelity study of the regenerated plants were analysed with 18 ISSR markers. A total of 159 monomorphic bands were obtained; one ISSR marker HB-12 showed a polymorphic band among the callus regenerants. The specific polymorphic fragment was then gel eluted, cloned and sequenced. Keywords: Rauvolfia serpentina, ISSR, Somaclonal variation, Medicinal plant, India

  • Cytogenetic variation appears to be more frequent in callus cultures than in other culture types, because callus formation implies a dedifferentiation phase followed by uncontrolled cell division.

“This cytogenetic variation appears to be more frequent in callus cultures than in other culture types, because callus formation implies a dedifferentiation phase followed by an uncontrolled cell division [37,38].”

The Abstract (page not specified) reports an ISSR polymorphic band among callus regenerants. Our copy does not address whether cytogenetic variation is more frequent in callus cultures than in other culture types or attribute such a difference to dedifferentiation and uncontrolled cell division. Searches for those terms and for comparisons with direct organogenesis found no supporting passage. (LLM, conf=0.73)

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
