Abnormal regulation of this signaling pathway is linked to a variety of cardiac disease conditions, including hypertrophy, fibrosis, arrhythmias, and infarction. Recent studies on genetically modified cellular and animal models document a crucial role of Wnt/β-catenin signaling in the molecular pathogenesis of arrhythmogenic cardiomyopathy (AC), an inherited disease of intercalated discs, typically characterized by ventricular arrhythmias and progressive substitution of the myocardium with fibrofatty tissue. In this review, we summarize the conflicting published data regarding the Wnt/β-catenin signaling contribution to AC pathogenesis and we report the identification of a new potential therapeutic molecule that prevents myocyte injury and cardiac dysfunction due to desmosome mutations in vitro and in vivo by interfering in this signaling pathway. Finally, we underline the potential function of microRNAs, epigenetic regulatory RNA factors reported to participate in several pathological responses in heart tissue and in the Wnt signaling network, as important modulators of Wnt/β-catenin signaling transduction in AC. Elucidation of the precise regulatory mechanism of Wnt/β-catenin signaling in AC molecular pathogenesis could provide fundamental insights for new mechanism-based therapeutic strategy to delay the onset or progression of this cardiac disease.

Suppression of canonical Wnt/β-catenin signaling leads to ACM, prominently through competition with nuclear localization of JUP, rather than through β-catenin degradation.
“In a landmark study by Garcia-Gras E et al. [77], these authors demonstrated that suppression of canonical Wnt/β-catenin signaling leads to ACM, prominently through competition with nuclear localization of JUP, rather than through β-catenin degradation [77,79,80].”

In 'Wnt signaling in AC: conflicting evidences' (page unavailable), the review reports that nuclear translocation of JUP/plakoglobin suppresses Wnt/β-catenin signaling and interferes with β-catenin/Tcf activity. It does not establish that this mechanism predominates over β-catenin degradation; the same section describes other, conflicting mechanisms in AC. (LLM, conf=0.86)

“The loss of nuclear YAP in conjunction with Wnt/β-catenin suppression further drives adipogenesis and maladaptive fibrotic repair [79,80].”
From A Paradigm Shift: Arrhythmogenic Cardiomyopathy Is an Inflammatory Disease (2026) · cites this paper as [80]
The Introduction’s “Wnt signaling in AC: conflicting evidences” section links cytoplasmic retention of YAP and suppression of Wnt/β-catenin activity to enhanced heart adipogenesis. It does not link that combined mechanism to maladaptive fibrotic repair; the separate “cardiac disease and repair” section instead discusses fibrogenesis in connection with Wnt activation. “The binding of Wnt recruits the cytoplasmic protein dishevelled (Dvl) to the cell membrane, thus sequestering the rate-limiting components axin and GSK3β from the degradation complex and determining the destruction of the complex [36, 37].”

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
