Abstract

Significance: The role of signal transducer and activator of transcription 3 (STAT3) pathway in hepatic stellate cells (HSCs) remains unclear. Using sorafenib and its derivative SC-1, we demonstrated the significant role of STAT3 pathway in liver fibrogenesis. We further found that STAT3 pathway of HSCs overexpressed in chronic hepatitis B patients with advanced fibrosis, therefore STAT3 may serve as a promising fibrotic biomarker and target. Furthermore, SHP-1 phosphatase-direct STAT3 inhibition may represent a previously unidentified strategy for antifibrotic drug discovery.

  • Increased tyrosine 705 phosphorylation of STAT3 in liver fibrosis can promote HSC proliferation and activation and contribute to liver fibrogenesis.

“Previous researchers have observed increased tyrosine 705 (Tyr 705) phosphorylation of STAT3 in liver fibrosis, which can promote HSC proliferation and activation and contribute to liver fibrogenesis [21, 22].”

The ‘P-STAT3 Overexpression in CHB Patients with Advanced Liver Fibrosis’ section reports increased p-STAT3 in HSCs as fibrosis advances in patients with chronic hepatitis B. The Discussion states that STAT3 activation in HSCs stimulates their proliferation and activation, but the passages identify p-STAT3 without specifying phosphorylation at Tyr 705; the patient finding is also limited to chronic hepatitis B. About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
