Abstract

This comprehensive review delves into the multifaceted roles of mesenchymal stem cells (MSCs) in leukemia, focusing on their interactions within the bone marrow microenvironment and their impact on leukemia pathogenesis, progression, and treatment resistance. MSCs, characterized by their ability to differentiate into various cell types and modulate the immune system, are integral to the BM niche, influencing hematopoietic stem cell maintenance and functionality. This review extensively explores the intricate relationship between MSCs and leukemic cells in acute myeloid leukemia, acute lymphoblastic leukemia, chronic myeloid leukemia, and chronic lymphocytic leukemia. This review also addresses the potential clinical applications of MSCs in leukemia treatment. MSCs’ role in hematopoietic stem cell transplantation, their antitumor effects, and strategies to disrupt chemo-resistance are discussed. Despite their therapeutic potential, the dual nature of MSCs in promoting and inhibiting tumor growth poses significant challenges. Further research is needed to understand MSCs’ biological mechanisms in hematologic malignancies and develop targeted therapeutic strategies. This in-depth exploration of MSCs in leukemia provides crucial insights for advancing treatment modalities and improving patient outcomes in hematologic malignancies.

  • Mesenchymal stromal cells may unintentionally facilitate tumor progression, especially in hematologic malignancies such as multiple myeloma.

“Multiple studies have shown that MSCs may unintentionally facilitate tumor progression, especially in hematologic malignancies such as MM, where the bone marrow microenvironment is already affected by cancerous plasma cells.”

From Mesenchymal stromal cells in bone marrow niche of patients with multiple myeloma: a double-edged sword (2025) · cites this paper as [297]

The Conclusions (section 10; page unavailable) say MSCs can aid tumor blood vessel formation, metastasis, relapse, and chemotherapy resistance in hematologic malignancies. The paper does not establish this specifically for multiple myeloma or describe a bone marrow microenvironment affected by cancerous plasma cells; its only exact mention of multiple myeloma is in a reference to a treatment study. • Interactions between mesenchymal stromal cells and malignant plasma cells in multiple myeloma can create a more supportive tumor microenvironment, potentially complicating treatment outcomes and disease prognosis.

“The interactions between mesenchymal stem cells and malignant plasma cells in multiple myeloma can create a more supportive tumor microenvironment, potentially complicating treatment outcomes and disease prognosis [297].”

From Mesenchymal stromal cells in bone marrow niche of patients with multiple myeloma: a double-edged sword (2025) · cites this paper as [297]

Section 8.1 (MSCs in CLL Progression; page unavailable) says interactions between mesenchymal stem cells and CLL cells create a supportive microenvironment and can influence treatment responses. The Conclusions (page unavailable) also discuss MSC-mediated drug resistance in hematologic malignancies. Neither passage establishes the claim specifically for malignant plasma cells in multiple myeloma, so the disease-specific attribution is unsupported. (LLM, conf=0.84)

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
