Decreased plasma β‐amyloid in the Alzheimer's disease APP A 673 T variant carriers
Abstract
We investigated the association of Alzheimer's disease (AD)-related rare variants APP A673T and ABCA7 rs200538373-C with the levels of β-amyloid (Aβ) and parameters of metabolic and cardiovascular health in a population-based cohort of healthy middle-aged and elderly men. Carriers of protective APP A673T variant had, on average, 28% lower levels of Aβ40 and Aβ42 in plasma as compared to the controls and the carriers of ABCA7 rs200538373-C. This is the first report to show decreased Aβ levels in plasma in APP A673T carriers and thus provides evidence that lower Aβ levels throughout life may be protective against AD. The E682K APP mutation favors Aβ production by suppressing β' cleavage and favoring β cleavage.
“In addition, the E682K mutation in APP, located at the P1, β site, favors Aβ production by suppressing the cleavage at the β’ site to favor cleavage at the β site (12,13).”

The A673T mutation is associated with 20% to 30% lower soluble APPβ levels compared with controls.
“In particular, the A673T mutation is associated with a different BACE1 recognition motif at the position P2 (A673T), resulting in 20% to 30% lower soluble APPβ (sAPPβ) levels compared with control subjects (12,13).”
From The β-Secretase BACE1 in Alzheimer’s Disease (2021) · cites this paper as [13]
Human carriers of the A673T variant have about 28% lower plasma Aβ40 and Aβ42 levels than controls.
“carriers of the A673T variant in humans have about 28% lower levels of Aβ40 and Aβ42 in plasma compared with control subjects (12,13).”

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
