MAO-B Inhibitors Do Not Block In Vivo Flortaucipir([18F]-AV-1451) Binding
Open accessAbstract
PURPOSE: F]AV-1451), we previously reported that non-demented Parkinson's disease patients show off-target binding in subcortical structures, but no appreciable cortical uptake. However, 59 % of these patients were receiving MAO-B inhibitors at the time of their scan. Here, we retrospectively investigated if MAO-B inhibitors in clinical doses affect flortaucipir binding. PROCEDURES: We compared the standard uptake values of flortaucipir at regional and voxel levels in Parkinson's disease patients who received MAO-B inhibitors with those who did not. RESULTS: Sixteen of 27 Parkinson's disease patients received MAO-B inhibitors at the time of scan. We found no significant flortaucipir uptake differences between the groups at voxel or regional levels. CONCLUSION: Use of MAO-B inhibitors at pharmaceutical levels did not significantly affect flortaucipir binding. Thus, MAO-B does not appear to be a significant binding target of flortaucipir.

The affinity of [18F]AV-1451 for monoamine oxidase is weak, and MAO inhibitors do not significantly displace its binding in patients with tauopathies.
“Furthermore, affinity of [18F]AV-1451 for monoamine oxidase is weak and the use of MAO-inhibitors does not significantly displace [18F]AV-1451 binding in patients with tauopathies37.”

“Binding to monoamine oxidase type B (MAO-B) has been raised, but recent evidence did not support MAO-B as a target.(49) It would also not be expected that MAO-B increases over time in subcortical regions of neurodegeneration.”

“Furthermore, unlike other tau PET tracers, 18F-AV-1451 is not appreciably blocked by monoamine oxidase B inhibitors (44), ruling out the possibility that the increased 18F-AV-1451 signal may simply correspond to an increase in monoamine oxidase B induced by the pathologic process (45).”

“Previous studies suggested that THK5351 has high affinity for MAO-B, as ingestion of MAO-B inhibitor (selegiline) reduced THK5351 uptake [2], whereas MAO-B inhibitor did not block flortaucipir uptake in human brains [13].”

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
