Abstract

The abnormal autophagy is associated with a variety of cardiovascular diseases. Long noncoding RNAs (lncRNAs) are emerging as new factors in gene regulation, but how lncRNAs operate in the regulation of autophagy in the heart is unclear. Here we report that a long noncoding RNA, named autophagy promoting factor (APF), can regulate autophagic cell death by targeting miR-188-3p and ATG7. The results show that miR-188-3p suppresses autophagy and myocardial infarction by targeting ATG7. Further, we find that APF lncRNA regulates miR-188-3p, and thus affects ATG7 expression, autophagic cell death and myocardial infarction. Our present study reveals a novel regulating model of autophagic programme, which comprises APF, miR-188-3p and ATG7 in the heart. Modulation of their levels may serve as potential targets and diagnostic tools for novel therapeutic strategies of myocardial infarction and heart failure. ATG7 promotes cardiomyocyte autophagy but the molecular mechanism of its regulation is unknown. Here, Wang et al.identify a long non-coding RNA dubbed autophagy promoting factor (APF) that binds and inhibits miR-188-3p, which in turn acts on ATG7, to regulate cardiac autophagy.

  • lncRNA APF is implicated in myocardial infarction pathogenesis by indirectly promoting ATG7 expression.

“Finally, the long non-coding RNA autophagy-promoting factor (APF) has been implicated in the pathogenesis of myocardial infarction owing to its ability to indirectly promote the expression of ATG7 [850].”
  • AAV-mediated deletion of Atg7 with Mir188-3p reduces size of myocardial infarction/area.

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
