Abstract

Age related macular degeneration (AMD) is a common retina-related disease leading to blindness. Little is known on the origin of the disease, but it is well documented that oxidative stress generated in the retinal pigment epithelium and choroid neovascularization are closely involved. The study of circulating miRNAs is opening new possibilities in terms of diagnosis and therapeutics. miRNAs can travel associated to lipoproteins or inside small Extracellular Vesicles (sEVs). A number of reports indicate a significant deregulation of circulating miRNAs in AMD and experimental approaches, but it is unclear whether sEVs present a significant miRNA cargo. The present work studies miRNA expression changes in sEVs released from ARPE-19 cells under oxidative conditions (i.e. hydrogen peroxide, H 2 O 2 ). H 2 O 2 increased sEVs release from ARPE-19 cells. Moreover, 218 miRNAs could be detected in control and H 2 O 2 induced-sEVs. Interestingly, only two of them (hsa-miR-302a and hsa-miR-122) were significantly under-expressed in H 2 O 2 -induced sEVs. Results herein suggest that the down regulation of miRNAs 302a and 122 might be related with previous studies showing sEVs-induced neovascularization after oxidative challenge in ARPE-19 cells.

  • miR-122-5p can suppress pro-inflammatory cytokine production and oxidative stress by targeting MAPK pathway components, including ERK and JNK.

“Previous studies have reported that miR-122-5p can suppress pro-inflammatory cytokine production and oxidative stress by targeting MAPK pathway components, including ERK and JNK 30 .”

The Discussion (page unavailable) reports that pre-miR-122 protects against H2O2-induced oxidative stress. It does not report suppression of pro-inflammatory cytokine production or identify ERK, JNK, or other MAPK components as targets; it instead names mitochondrial ribosomal protein S11. • The three selected miRNAs-hsa-miR-122-5p, hsa-let-7a-5p, and hsa-miR-26a-5p-provide complementary and comprehensive coverage of almost all critical genes associated with the MAPK signaling pathway.

“From this analysis, we selected the three abundant miRNAs-hsa-miR-122-5p, hsa-let-7a-5p, and hsa-miR26a-5p-based on their complementary and comprehensive coverage of almost all the critical genes associated with the MAPK signaling pathway 27-33 as analyzed using miRTarBase version 8.0 and TarBase v8.0 integrated within miRNet (Figure S1).”

From rAAV-mediated Multi-miRNA Delivery Targeting MAPK Pathway Suppresses Laser-induced Choroidal Neovascularization in Mice (2026) · cites this paper as [30]

The Results section on pathway analysis discusses let-7a and miR-122, but does not say that they cover critical MAPK genes. Searches for MAPK, miR-26a-5p, and comprehensive gene coverage found no passage supporting the three-miRNA coverage claim; the paper instead describes other pathways and uses different target-prediction tools. About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
