Abstract

In patients with fibrodysplasia ossificans progressiva, the disease-causing mutation changes the ligand response profile of the ACVR1 receptor so that it is activated by the normally antagonistic ligand activin A.

  • ActRIIA/B receptors mediate many diverse biologic processes by responding to over a dozen ligands.

“as ActRIIA/B receptors mediate many diverse biologic processes by responding to over a dozen ligands23–25”

From GDF8 and activin A blockade protects against GLP-1–induced muscle loss while enhancing fat loss in obese male mice and non-human primates (2025) · cites this paper as [24]

The Results section on ligand-dependent heterotopic ossification describes ACVR2A and ACVR2B as interacting with both BMPs and activins, supporting a broad ligand repertoire. The Results section on activin A also describes their participation in complexes involving several activins. The paper does not establish that these receptors respond to over a dozen ligands or that they mediate many diverse biological processes. (LLM, conf=0.82)

  • There are potential concerns with broad ActRIIA/B blockade as an approach to maintain or increase muscle growth because these receptors mediate diverse biologic actions of more than a dozen ligands.

“since these receptors mediate the diverse biologic actions of more than a dozen ligands23–25, there are potential concerns with broad ActRIIA/B blockade as an approach to maintain or increase muscle growth.”

From GDF8 and activin A blockade protects against GLP-1–induced muscle loss while enhancing fat loss in obese male mice and non-human primates (2025) · cites this paper as [24]

The Results section (“ACVR1[R206H]-driven HO is a ligand-dependent process”; page unavailable) describes ACVR2A-Fc and ACVR2B-Fc as broad-acting blockers of BMPs and activins. The paper thus supports the premise that these receptors are involved with multiple ligands, but it does not establish “more than a dozen” ligands or assess broad blockade as an approach to maintain or increase muscle growth. The potential concern is the citing authors’ inference, not a conclusion this source makes. (LLM, conf=0.82)

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
