A review on formulation and analytical aspect of mangiferin a natural phenolic xanthonoid
Open accessAbstract
Recently there has been increased interest in nutraceuticals because of a high safety margin, significant role in treating number of diseases and minimal probability of adverse reactions. Nevertheless, certain obstacles, such as low permeability and poor solubility, which lead to low bioavailability, limit their effectiveness. The therapeutic effect of nutraceuticals has improved as a result of delivery carriers' demonstrated ability to get around nearly all of the aforementioned constraints. Mangiferin, a polyphenol from the mango tree that has many positive pharmacological actions, is one of the promising nutraceuticals that has attracted a lot of interest recently. However, its solubility in aqueous phase is very low and other issues that limit its efficacy and bioavailability. The pharmacological actions of mangiferin are outlined in this review, which also offers insight into how delivery vehicles (Complexes, protein nanoparticles, inorganic nanoparticles, polymeric nanoparticles, vesicular nanoparticles, and lipidic nanoparticles) can overcome mangiferin pharmaceutical challenges, thereby improving its therapeutic effects in the treatment of various diseases. The analytical methodologies available for the estimation of this natural product in various matrix was also discussed in this review.

Low solubility and permeability constrain Cmax/AUC after oral dosing.
“i) Low solubility and permeability [constraining Cmax/area under the curve (AUC) after oral dosing] ((164-166)”
From Mangiferin in human disease: Multifaceted mechanisms and applications (Review) (2026) · cites this paper as [165]
Section 1.2.1 (Bioavailability; page unavailable) says mangiferin’s low solubility and poor permeability result in low oral bioavailability. Section 2.6 (Complexes; page unavailable) reports a higher Cmax with a formulation that improves solubility and permeability, but the copy does not explicitly link these limitations to AUC after oral dosing. (LLM, conf=0.82)
Experimental — accuracy not yet measuredMicrobiome-dependent deglycosylation introduces variability.
“iii) variability introduced by microbiome-dependent deglycosylation (165,169)”
From Mangiferin in human disease: Multifaceted mechanisms and applications (Review) (2026) · cites this paper as [165]
Section 1.2.3 (Metabolism; page not available) identifies deglycosylation as an initial step in mangiferin metabolism. The Review passage also says gut flora facilitates mangiferin absorption and metabolism, but the copy does not specifically say that deglycosylation depends on the microbiome or that this process introduces variability. (LLM, conf=0.83)
Experimental — accuracy not yet measured

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
