Abstract

The response to DNA damage is critical for cellular homeostasis, tumor suppression, immunity, and gametogenesis. In order to provide an unbiased and global view of the DNA damage response in human cells, we undertook 31 CRISPR-Cas9 screens against 27 genotoxic agents in the retinal pigment epithelium-1 (RPE1) cell line. These screens identified 890 genes whose loss causes either sensitivity or resistance to DNA-damaging agents. Mining this dataset, we discovered that ERCC6L2 (which is mutated in a bone-marrow failure syndrome) codes for a canonical non-homologous end-joining pathway factor, that the RNA polymerase II component ELOF1 modulates the response to transcription-blocking agents, and that the cytotoxicity of the G-quadruplex ligand pyridostatin involves trapping topoisomerase II on DNA. This map of the DNA damage response provides a rich resource to study this fundamental cellular system and has implications for the development and use of genotoxic agents in cancer therapy.

  • Many genes identified from iCRISPRi pooled screens in 3D human gastric organoids were not determined as cisplatin sensitization partners in previous genome-scale CRISPR KO screens in immortalized 2D human cell lines.

“In particular, many genes identified from our iCRISPRi pooled screens in 3D human gastric organoids were not determined as cisplatin sensitization partners in previous genome-scale CRISPR KO screens in immortalized 2D human cell lines51, suggesting potential 3D specificity (Fig. S6A).”
  • Recent genome-wide investigations underscore that these effects are not confined to promotors, but are rather observed across the genome.

“Recent genome-wide investigations underscore that these effects are not confined to promotors, but are rather observed across the genome (68–70).”

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
