Abstract

To investigate circuit mechanisms underlying locomotor behavior, we used serial-section electron microscopy (EM) to acquire a synapse-resolution dataset containing the ventral nerve cord (VNC) of an adult female Drosophila melanogaster. To generate this dataset, we developed GridTape, a technology that combines automated serial-section collection with automated high-throughput transmission EM. Using this dataset, we studied neuronal networks that control leg and wing movements by reconstructing all 507 motor neurons that control the limbs. We show that a specific class of leg sensory neurons synapses directly onto motor neurons with the largest-caliber axons on both sides of the body, representing a unique pathway for fast limb control. We provide open access to the dataset and reconstructions registered to a standard atlas to permit matching of cells between EM and light microscopy data. We also provide GridTape instrumentation designs and software to make large-scale EM more accessible and affordable to the scientific community.

Claims other papers make about this one

Each is checked against this paper’s own text: does it say what the citing paper says it does?

  • Intrinsic neurons of the brain make up three-quarters of the adult fly nervous system.

“Intrinsic neurons of the brain make up three-quarters of the adult fly nervous system54–56 and amount to 85% of brain neurons.”

From Neuronal wiring diagram of an adult brain (2024) · cites this paper as [54]

The Discussion section, “A synapse-resolution adult Drosophila VNC dataset” (page unavailable), says the VNC contains about a third of the neurons in the adult central nervous system. That leaves about two-thirds for the brain, including its intrinsic neurons; the paper does not support their making up three-quarters of the larger, whole nervous system. (LLM, conf=0.82)

  • The adult fruit fly has only ∼ 200,000 neurons in its brain, compared with ∼ 70 million in the mouse brain and ∼ 80 billion in the human brain.

“It has only ∼ 200,000 neurons in its brain 24 (compared with ∼ 70 million in the mouse brain 25 and ∼ 80 billion in the human brain 26 ).”

From NeuroMechFly v2: simulating embodied sensorimotor control in adult Drosophila (2024) · cites this paper as [25]

The Introduction describes the Drosophila nervous system as small, and the Discussion ('A synapse-resolution adult Drosophila VNC dataset') says the VNC contains about a third of the neurons in the adult central nervous system. Neither gives a count of neurons in the adult fly brain; searches for '200,000' and brain-neuron counts found no supporting passage. The mouse and human figures have separate citations in the citing sentence and are not judged here. (LLM, conf=0.76)

  • Leg muscles are estimated to be innervated by around 70 MNs in each leg, that originate from ~15 hemilineages.

“These leg muscles are estimated to be innervated by around 70 MNs in each leg, that originate from ~15 hemilineages (Baek and Mann, 2009; Brierley et al., 2012; Phelps et al., 2021).”
  • The FANC is the first complete EM volume of the adult Drosophila VNC, providing valuable insight into MN morphology, count and organization in the VNC thoracic segments.

“Of note, the FANC (Phelps et al., 2021) is the first complete EM volume of the adult Drosophila VNC, providing valuable insight into MN morphology, count and organization in the VNC thoracic segments, as well as allowing labs to further reconstruct and investigate VNC circuits of interest.”
  • These numbers were similar to those from the FANC dataset, where MNs traced (excluding AbN2-4 and AbNT) numbered 507; we found 510 for the MNs in the same nerves.

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
