Abstract

Polydeoxyribonucleotide (PDRN) consists of DNA fragments with molecular weights ranging from 50 to 1 500 kDa, which are mainly extracted from the sperm cells of salmon trout or chum salmon. Many preclinical and clinical studies have demonstrated the properties of PDRN. These include anti-inflammatory, anti-apoptotic, anti-osteoporotic, anti-melanogenetic, anti-allodynic, anti-osteonecrotic, bone regenerative, tissue damage preventive, anti-ulcerative, and wound healing properties, which are mediated by the activation of the adenosine A2A receptor and salvage pathways. Moreover, PDRN promotes angiogenesis, cellular activity, collagen synthesis, soft tissue regeneration, and skin priming and revitalization and can be used to treat hyperpigmentation. Therefore, this review assessed the potential use of PDRN as an anti-aging agent for the skin.

  • Both PN and PDRN are nucleic acid-derived polymers composed of nucleotide monomers connected via phosphodiester bonds.

“Both PN and PDRN are nucleic acid-derived polymers composed of nucleotide monomers connected via phosphodiester bonds [11,17].”

From Comparison of Polynucleotide and Polydeoxyribonucleotide in Dermatology: Molecular Mechanisms and Clinical Perspectives (2025) · cites this paper as [17]

The Introduction states that PDRN is a DNA-derived linear polymer of deoxyribonucleotide monomers joined by phosphodiester linkages. The paper also mentions polynucleotide-containing products, but the passages found do not separately describe PN’s composition or linkages, so the claim about both is only partly supported. (LLM, conf=0.82)

  • PDRN has demonstrated a strong safety profile in both acute and chronic toxicity studies, with no significant immunogenic responses.

“PDRN has demonstrated a strong safety profile in both acute and chronic toxicity studies, where no significant immunogenic responses were observed [11,17,19].”

From Comparison of Polynucleotide and Polydeoxyribonucleotide in Dermatology: Molecular Mechanisms and Clinical Perspectives (2025) · cites this paper as [17]

The “Safety of use” passage reports no toxicity after repeated systemic administration in mice and rats and attributes an absence of immunological effects to PDRN’s purification process. It supports a favorable safety profile, but does not describe both acute and chronic toxicity studies or report measured immunogenic responses in those studies. (LLM, conf=0.87)

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
