The seven faces of SIRT7
Open accessAbstract
-dependent protein deacetylases, is a key mediator of many cellular activities. SIRT7 expression is linked to cell proliferation and oncogenic activity, connecting SIRT7-dependent regulation of ribosome biogenesis with checkpoints controlling cell cycle progression, metabolic homeostasis, stress resistance, aging and tumorigenesis. Despite this important functional link, the enzymatic activity, the molecular targets and physiological functions of SIRT7 are poorly defined. Here, we review recent progress in SIRT7 research and elaborate the main pathways in which SIRT7 participates.

SIRT7 appears necessary for preserving active rDNA transcription under conditions of cellular growth and stress.
“In contrast to the broadly repressive actions often attributed to other sirtuins, SIRT7 appears necessary for preserving active rDNA transcription under conditions of cellular growth and stress [183,184].”
From Ribosome Biogenesis as a Putative Bottleneck to Skeletal Muscle Hypertrophy: Mechanisms, Human Evidence, and Practical Modulators (2026) · cites this paper as [184]
The section “SIRT7 promotes pre-rRNA transcription and processing” says SIRT7 facilitates rDNA transcription and links its activity to cell proliferation. But “SIRT7 attenuates cellular stress” says osmotic or transcriptional stress releases SIRT7 from nucleoli and downregulates Pol I transcription, rather than preserving active rDNA transcription during stress. Page numbers are unavailable in our copy. (LLM, conf=0.88)
Experimental — accuracy not yet measuredGiven its dependence on intracellular NAD+ availability, SIRT7 may provide a mechanistic link between cellular energetic status and nucleolar function.

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
