Lactate is a Preferential Oxidative Energy Substrate over Glucose for Neurons in Culture
Open accessAbstract
The authors investigated concomitant lactate and glucose metabolism in primary neuronal cultures using 13 C- and 1 H-NMR spectroscopy. Neurons were incubated in a medium containing either [1- 13 C]glucose and different unlabeled lactate concentrations, or unlabeled glucose and different [3- 13 C]lactate concentrations. Overall, 13 C-NMR spectra of cellular extracts showed that more 13 C was incorporated into glutamate when lactate was the enriched substrate. Glutamate 13 C-enrichment was also found to be much higher in lactate-labeled than in glucose-labeled conditions. When glucose and lactate concentrations were identical (5.5 mmol/L), relative contributions of glucose and lactate to neuronal oxidative metabolism amounted to 21% and 79%, respectively. Results clearly indicate that when neurons are in the presence of both glucose and lactate, they preferentially use lactate as their main oxidative substrate.

The original ANLS has been controversially discussed but has also gained considerable experimental support by in vitro and in vivo studies.
“While the original ANLS has been controversially discussed (Albanese et al., 2016;Bosoi & Rose, 2014;Dienel, 2017;Liguori et al., 2015;Yellen, 2018), it has also gained considerable experimental support by in vitro (Bouzier-Sore et al., 2003;Karagiannis et al., 2021) and in vivo studies (Mächler et al., 2016;Suzuki et al., 2011;Wyss et al., 2011;Zuend et al., 2020).”

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
