Mechanisms underlying microbial-mediated changes in social behavior in mouse models of autism spectrum disorder
Open accessAbstract
SUMMARY Currently, there are no medications that effectively treat the core symptoms of Autism Spectrum Disorder (ASD). We recently found that the bacterial species Lactobacillus (L.) reuteri reverses social deficits in maternal high-fat diet offspring. However, whether the effect of L. reuteri on social behavior is generalizable to other ASD models and its mechanism(s) of action remain unknown. Here, we found that treatment with L. reuteri selectively rescues social deficits in genetic, environmental, and idiopathic ASD models. Interestingly, the effects of L. reuteri on social behavior are not mediated by restoring the composition of the host’s microbiome, which is altered in all of these ASD models. Instead, L. reuteri acts in a vagus nerve-dependent manner and rescues social interaction-induced synaptic plasticity in the ventral tegmental area of ASD mice, but not in oxytocin receptor-deficient mice. Collectively, treatment with L. reuteri emerges as promising non-invasive microbial-based avenue to combat ASD-related social dysfunction.

TL;DR
Treatment with *Lactobacillus reuteri* selectively rescues social deficits across genetic, environmental, and idiopathic mouse models of ASD, acting independently of overall microbiome composition but requiring the vagus nerve and oxytocin receptor signaling in dopaminergic neurons. The rescue is associated with restored social interaction-induced synaptic plasticity in ventral tegmental area dopamine neurons.
Key findings
- *L. reuteri* treatment (~1×10⁸ organisms/mouse/day for 4 weeks) reversed social deficits in Shank3B−/−, VPA, BTBR, and germ-free mice; effects were selective for social behavior, as hypoactivity in Shank3B−/− mice was not improved.
- *L. reuteri* did not significantly alter overall gut microbial composition in Shank3B−/− or BTBR mice, and monocolonization reversed social deficits in germ-free mice, indicating it acts without other microbes.
- Bilateral subdiaphragmatic vagotomy prevented *L. reuteri* rescue in Shank3B−/− mice, whereas sham-operated mice were rescued; intranasal oxytocin restored social behavior in vagotomized Shank3B−/− mice.
- *L. reuteri* and oxytocin failed to rescue social deficits or VTA plasticity in DA-Oxtr−/− mice, and the oxytocin receptor antagonist L-371,257 prevented *L. reuteri*’s prosocial effects; Shank3B−/− mice had reduced oxytocin-positive PVN neurons, which *L. reuteri* increased.
Methods
- Mouse models: C57BL/6J, Shank3B−/−, VPA-exposed, BTBR, germ-free, and DA-Oxtr−/− mice; only male mice were included, with at least two litters per experiment.
- Behavioral assays: three-chamber sociability and social novelty, reciprocal social interaction, open field, and buried food tests; 16S rRNA gene sequencing and qPCR for *L. reuteri*; FITC-dextran intestinal permeability assay; RT-qPCR for tight junction genes.
- Mechanistic assays: bilateral subdiaphragmatic vagotomy with CCK-8 satiety verification; intranasal oxytocin (200 μg/kg) and L-371,257 (~300 μg/kg); immunofluorescence for oxytocin/NeuN in the PVN; whole-cell electrophysiology measuring AMPAR/NMDAR ratios in VTA dopamine neurons after social interaction or cocaine.
Limitations
- Stated: how precisely *L. reuteri* interacts with the vagus nerve remains unknown; future studies are needed in non-mouse models and human clinical trials, and safety/long-lasting effects in humans are not established.
- Stated/evident: only male mice were included, so sex differences are not addressed.
- Evident: many behavioral and microbiome results rely on supplementary figures, and the main text does not report exact group sizes or effect sizes; translation to human ASD is not tested.

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Claims read by deepseek-v4.1-flash under definition version 3 (extract-v3) on 2026-10-10.
Classified by jev-1.13-20260917 (gold-guideline-v0.2) on 2026-10-10.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
