Abstract

Opening and closing blood enhancers: As cells develop and differentiate into different types, the shape and accessibility of their DNA can change. Lara-Astiaso et al. studied this phenomenon in blood. They developed a technique that examines a relatively small number of cells to identify the changes that affect DNA during blood development. They found that the DNA of noncoding regions, called enhancers, is set in an open position when cells are undifferentiated and able to take on a variety of roles and gradually closes as cells mature into their final forms. • The majority of up- and downregulated genes upon vitC treatment are associated with one or more haematopoietic enhancers within 20 kb of their transcription start sites.

“The majority of up- (373/450; 83%) and downregulated (165/221; 75%) genes are associated with one or more haematopoietic enhancers37 within 20 kb of their transcription start sites.”
  • A significant fraction of IDH1R132H cell enhancers overlapped the catalogue of haematopoietic enhancers.

“A significant fraction of IDH1R132H cell enhancers overlapped the catalogue of haematopoietic enhancers (1358/4389 (30%) and 1276/3650 (35%) for untreated and vitC-treated, respectively).”
  • Intersection of Gfi1 and Irf8 peaks revealed shared regions deemed accessible in GMPs based on ATAC-Seq.

“Intersection of the Gfi1 and Irf8 peaks revealed shared regions that were deemed accessible in GMPs27 based on ATAC-Seq27 (Fig. 2b, c).”

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
