Comprehensive Multi-omics Analysis Reveals Mitochondrial Stress as a Central Biological Hub for Spaceflight Impact
Open accessAbstract
Summary Spaceflight is known to impose changes on human physiology with unknown molecular etiologies. To reveal these causes, we used a multi-omics, systems biology analytical approach using biomedical profiles from fifty-nine astronauts and data from NASA’s GeneLab derived from hundreds of samples flown in space, to determine transcriptomic, proteomic, metabolomic, and epigenetic responses to spaceflight. Overall pathway analyses on the multi-omic datasets showed significant enrichment for mitochondrial processes, as well as innate immunity, chronic inflammation, cell cycle, circadian rhythm, and olfactory functions. Importantly, NASA’s Twin Study provided a platform to confirm several of our principal findings. Evidence of altered mitochondrial function and DNA damage was also found in the urine and blood metabolic data compiled from the astronaut cohort and NASA Twin Study data, indicating mitochondrial stress as a consistent phenotype of spaceflight.

Re-establishing mitochondrial function will also recuperate the proteostasis condition.

The Summary (page unavailable) reports the opposite intervention in C. elegans: reduced mitochondrial electron transport chain function during development protects against age-related proteostasis collapse. The Results likewise attribute protection against cytosolic protein aggregation to reduced ETC function, not to re-establishing mitochondrial function. (LLM, conf=0.84)

Spaceflight induces proteostasis failure.

The Abstract and ‘An Intimate Connection between Mitochondrial Function and Cytosolic Proteostasis’ section link mitochondrial dysfunction and disrupted translation or ribosome quality control to proteostasis failure (page numbers unavailable). Reduced muscle loading due to microgravity is a strong driver of gene regulation that can influence transcriptional responses to spaceflight.

The Results section on rodent housing conditions reports differential gene expression in three types of muscle, so the paper supports the general point that muscle transcription responds to an environmental condition relevant to spaceflight. But these were comparisons between ground-control housing types; the paper does not identify reduced muscle loading from microgravity as a strong driver of those changes. The Discussion (p. 10) links the ATF family to the AARE response and identifies ATF5 as an early responder to mitochondrial stress. But the results on dividing cells (p. 5) identify ATF4, not ATF5, as an upstream regulator of one-carbon metabolism genes, FGF21, and GDF15; they also say ATF5 silencing was not essential for induction of that response. The copy does not show ATF5 binding amino acid response elements or directly upregulating the listed pathways and factors. (LLM, conf=0.86)

About these checks
Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.
Claims read by deepseek-v4.1-flash under definition version 3 (extract-v3) on 2026-09-28.
Classified by jev-1.13-20260917 (gold-guideline-v0.2) on 2026-09-28.
Cited sources are checked by gpt-6-sol.
Statistics and arithmetic are recomputed, not judged.
Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
