Abstract

Over the past two decades, the molecular machinery that underlies autophagic responses has been characterized with ever increasing precision in multiple model organisms. Moreover, it has become clear that autophagy and autophagy-related processes have profound implications for human pathophysiology. However, considerable confusion persists about the use of appropriate terms to indicate specific types of autophagy and some components of the autophagy machinery, which may have detrimental effects on the expansion of the field. Driven by the overt recognition of such a potential obstacle, a panel of leading experts in the field attempts here to define several autophagy-related terms based on specific biochemical features. The ultimate objective of this collaborative exchange is to formulate recommendations that facilitate the dissemination of knowledge within and outside the field of autophagy research.

  • OPTN actively regulates proteasomal turnover of RIPK1, as loss of OPTN induces axonal degeneration via RIPK1-dependent necroptosis.

“Moreover, it has recently been reported that the autophagic receptor optineurin (OPTN) [509] actively regulates the proteasomal turnover of RIPK1, as the loss of OPTN induces axonal degeneration via RIPK1-dependent necroptosis [510].”
  • Once engulfed, entotic cells are often eliminated by a RCD subroutine independent of BCL2 proteins and caspases, but relying at least in part on LC3-associated phagocytosis.

“Once engulfed, entotic cells are often eliminated by a RCD subroutine that occurs independently of BCL2 proteins and caspases [694, 704], but relies (at least in part) on a specific autophagy-related process commonly known as LC3-associated phagocytosis (LAP) [509, 705, 706].”
  • LC3 has a known role in autophagy and has been recently identified as a regulator of rapid apoptotic cell clearance in macrophages in LC3-associated phagocytosis, which occurs in the absence of immune activation.

“In addition to its known role in autophagy22, LC3 has been recently identified as a novel regulator of rapid apoptotic cell clearance in macrophages in a process termed LC3-associated phagocytosis (LAP), which occurs in the absence of immune activation23.”

About these checks

Registry, retraction, Cites and statistics results are computed, not judged, with one exception: a printed reference that no index lists is read once by a model before it counts as a citation error. Claims are extracted by a model and kept only when their quote appears verbatim in the text; each cited source is then checked, with a model’s help, for whether it says what the claim relies on. Those verdicts can be wrong — read the claim’s quote and the cited source before relying on one.

Statistics and arithmetic are recomputed, not judged.

Confidence bands are shown only for fields calibrated on our annotated test set. None is calibrated yet, so no band is shown.
